Cyclins are proteins that activate cyclin-dependent kinases (CDKs).  CDK-cyclin complexes play diverse and crucial roles in the functioning of cells, often becoming dysregulated in cancers. This invited review for a special issue on the cyclin protein family summarises our current knowledge of cyclin structures. It highlights how structural knowledge can help us to understand their diverse functions and mechanisms of regulation. Finally, we look at ways to identify additional protein binding sites on cyclins and their partner proteins which may yield new targetable interactions for therapeutic intervention.
