Project Summary: This project aims to study human protein complexes associated with genetic diseases in their native environment. This will be done by utilising CRISPR genome editing technology to add affinity tags to endogenous proteins for purification. This method allows for copurification with interacting proteins which recombinant expression of the protein of interest does not. Structural analysis of the purified complex by cryo-EM will then allow characterisation of the interprotein interactions and identification of novel interactors. This will provide us with a better understanding of the underlying molecular mechanism of disease and potentially reveal therapeutic targets.
