Chemical Probes for Kinases, E3 Ligases and Beyond

Chemical probes, well-validated modulators of protein function, are essential tools for studying the biological and therapeutic relevance of proteins. The Structural Genomics Consortium (SGC), a public-private partnership, has long developed and shared these probes with the scientific community. Initially focused on epigenetic targets, the SGC has since expanded its efforts to generate specific probes for diverse proteins, including kinases and E3 ligases. More recently, we have expanded this work by assembling chemogenomic libraries, well-annotated, narrowly specific compound subsets that provide characterized tools for 1,000+ targets. To expand the target space further, PROTAC strategies are employed. Moving forward, we will focus on accelerating hit finding and chemical probe development by collaborating with the computational drug discovery community to generate the data needed to transform early drug discovery into a primarily computational approach, accelerating the discovery of new medicines supported by artificial intelligence (AI) and machine learning (ML) technology.
