  • A novel and efficient translational read-through generates a sorting receptor in Saccharomyces cerevisiae

The Sacchromyces cerivisiae genome encodes a fourth potential sorting receptor encoded by an mRNA which contains a premature stop codon followed by a sequence which could potentially base pair to form a stem loop structure. By placing different cloned constructs of this stem loop into yeast cultures, protein lysates from the cultures were used in a dual light assay to measure the percentage read-through of this stop codon during translation. The aim of this project was to then identify how much of this structure is required to be present for effective readthrough to produce the full translated protein.
